Destroy All Humans! is a complete remake of the first installment in the humorous action game series originally released in 2005. Players once again take on the role of Crypto, an alien wreaking havoc on Earth during the mid-20th century.

The story is set in the United States in 1959, where the protagonist, Cryptosporidium-137, a gray-skinned, large-eyed alien belonging to the Furon race, arrives on Earth to fulfill a mission assigned by his superior, Orthopox-13. The mission involves locating the missing Cryptosporidium-136, who was preparing plans for mass DNA extraction from human bodies. This DNA is crucial for the survival of the Furon race, which has lost its reproductive abilities and relies on cloning technology to avoid extinction. Humans possess fragments of Furon DNA in their genetic code, remnants of ancient visits by these extraterrestrials.

The gameplay is presented from a third-person perspective, with players controlling Crypto as he navigates through large, open 3D environments representing 1950s America. The game features both typical suburban areas and secret military bases. The primary objectives include eliminating humans and extracting their DNA, which is essential for progressing in the game.
